The two d35S fragments are as diagrammed in Figure 1. − and + indicate McrBC-untreated and treated genomic DNA, respectively. ‘H2O’ is a negative control PCR without genomic DNA. McrBC digests methylated DNA to result in reduced PCR product amounts. (B) Bisulfite sequencing analysis of cytosine methylation in d35S in LUCH in wild type and ago4–6. The top strand of d35S #3 in Figure 1 was analyzed. (C) d35S-specific siRNA accumulation in the LUCH line as detected by northern blotting. The numbers indicate the amount of enriched small RNAs loaded into the gel. Col-0, wild type (with no transgene).
